I took a job and the company has already started the work permit but I got a better offer with another company can the work permit be canceled or can they company have a transfer done.

You can cancel your current permit and start the process over with the new employer, as long as they have advertised the job and followed the procedure the Labour Department requires.
Once required a work permit, can I leave and re-enter within that year? or would I have to redo paperwork?
